作者王喆(2019)在《OsDLT基因调控的水稻抗虫防御信号网络解析》一文中研究指出:油菜素内酯(brassinosteroids,BRs)是一种多羟基甾醇类化合物,具有极高的生物活性是世界公认的第六大植物激素。其参与调控植物生长发育的许多过程,同时在植物响应生物和非生物胁迫中发挥作用。水稻(Oryza sativa)是世界上最重要的粮食作物之一,其产量和质量关乎绝大部分人口的生存,而褐飞虱(Nilaparvata lugens St?l,BPH)作为水稻单食性害虫是水稻生产中重要的害虫之一。本文从一个油菜素内酯信号通路下游的转录因子OsDLT(DWARF AND LOW-TILLERING)出发,研究其调控的水稻抗虫防御信号网络,结果如下:(1)相比于相同发育阶段的野生型水稻,突变体水稻dlt对褐飞虱的取食危害具有更强的耐受性。同时,不同发育阶段的野生型水稻和48日龄dlt水稻的褐飞虱选择性实验表明褐飞虱更喜爱危害各个发育阶段的野生型水稻。(2)OsDLT基因过量表达增加了水稻对褐飞虱的敏感性,与野生型水稻相比褐飞虱雌成虫更喜欢在过量表达OsDLT取食和产卵。取食过量表达OsDLT水稻的初羽化雌成虫分泌的蜜露量显著增加;褐飞虱若虫发育历期显著缩短,F1代单雌产卵量和卵孵化率显著上升。(3)与未处理9311相比,褐飞虱雌成虫更趋向于在24-表油菜素内酯(24-epicastasterone,ECS)处理的9311水稻上取食和产卵;而褐飞虱雌成虫对未处理和ECS处理的OsDLT功能确实突变体水稻dlt之间无取食和产卵偏好。丙环唑(Propiconazole,PCZ)是植物BR合成抑制剂,与未处理9311相比褐飞虱雌成虫在PCZ处理9311水稻上的取食和产卵量显著减少;而与未处理的dlt突变体水稻相比,褐飞虱雌成虫在PCZ处理的dlt水稻上的取食和产卵量显著增加。与未处理的9311水稻相比,经Bikinin处理的9311水稻对褐飞虱雌成虫取食和产卵的敏感性显著地降低;而dlt突变体,Bikinin处理增加褐飞虱雌成虫取食和产卵敏感性。(4)转录组数据分析后得到7196个差异表达基因。对褐飞虱取食处理9311水稻(WB)和dlt水稻(dB)出现的差异表达基因进行分析,茉莉酸途径中总共有18个基因出现了上调,占该途径差异表达基因的81.83%。水杨酸途径中总共有15个基因出现差异表达,其中11个基因出现上调占水杨酸途径差异表达基因的73.33%。18个防御相关的WRKY家族占WRKY家族差异表达基因的85.71%,9个ERF家族差异表达基因全部上调。3个病程相关蛋白蛋白、5个几丁质酶出现了显著上调。(5)通过构建酵母双杂交体系证明了OsDLT蛋白自身不具有自激活并筛选出3个和OsDLT蛋白具有相互作用的蛋白编码基因。之后克隆并构建了4个茉莉酸抑制子JAZ家族基因的激活质粒,通过酵母双杂交证明OsJAZ6与OsDLT蛋白可以产生相互作用。本文研究表明,OsDLT在褐飞虱诱导的防御反应中发挥重要作用,该基因负调控水稻对褐飞虱的抗性;OsDLT可能是油菜素内酯途径与茉莉酸途径互作的重要因子,在调控水稻的抗虫防御反应。
